Use As an Affinity Column

Advanced affinity columns can be fabricated by immobilizing Protein-A, which specifically interacts with IgG, onto SPM.

Compared with a commercially available protein A (Column-1) column packed with particles, the SPM column enables more efficient separation. In the particle-packed column, a large amount of analyte passes through at the reduced flow rate of a mobile phase, resulting in incomplete separation. In contrast, the SPM column can transfer solution at less than one-fifth pressure required for conventional columns. Even at flow rates more than ten times higher than the normal flow rate (1.0 mL/min), the SPM column maintains efficient separation selectivity and recovery.

Protein A–immobilized SPM

The chromatograms illustrate the performance of a Protein A–immobilized spongy monolith (ProA-SPM) at various flow rates (1–9 mL/min).

ProA-SPM column can be operated at flow rates more than three times higher than conventional particle-packed columns without compromising separation performance.

 

The chromatograms show a comparison between a ProA-SPM column and a commercially available ProA particle-packed column (ProA-Column).
In the ProA-Column, a significant flow-through peak is observed, indicating that some analytes pass through the column without being retained.

In contrast, the ProA-SPM column effectively retains IgG, resulting in a clear retained peak with minimal loss in the flow-through fraction.
Furthermore, the ProA-SPM column achieves low back pressure and a high recovery rate even at a high flow rate (9.0 mL/min).

Comparison of Column Pressure
