The use of affinity column

Advanced affinity columns can be fabricated by binding Protein-A, which specifically interacts with IgG, to the SPM.

In comparison with efficiency between protein A columns with packed particles and the SPM, when the flow rate of a mobile phase increased, some analytes pass through the column, resulting in incomplete separation. However, solution can be transferred in the SPM column with less than one fifth pressures compared to commercially available columns. Efficient separation selectivity and the high recovery rate were achieved even at more than ten times faster than the normal flow rate (1.0 mL/min).

Comparison of column pressure with soft sepharose particles

The SPM enables high flow rate (10 mL/ min) at under 4 MPa, while the immediate increase in pressure was observed at more than 4 mL/min flow rate with a sepharose 6B (45 – 165µm) column as particles are soft.
